Exopolysaccharide production by Paenibacillus sp. and detection of antifungal activity in the fraction remaining after EPS separation
Main Article Content
Abstract
This study investigated the growth of Paenibacillus sp. in various liquid nutrient media and the preliminary detection of fractions exhibiting antifungal activity against Fusarium oxysporum f. sp. vasinfectum. The bacterium was cultivated in Nutrient Broth (NB), Luria–Bertani (LB), Potato Dextrose (PD), Tryptone Soy Broth (TSB), ISP2 and Corn Steep Liquor (CSL) media at 28 °C and 150 rpm for 5 days. Preliminary testing of the resulting extracts revealed no significant antifungal effect. Exopolysaccharide (EPS) production was observed in ISP2 medium, and the EPS was separated using ethanol. When tested separately, the EPS fraction did not inhibit the growth of F. oxysporum f. sp. vasinfectum, whereas the fraction remaining after EPS separation suppressed the growth of the fungal mycelium. The results indicate that the active compounds need to be isolated by chromatographic methods and their chemical nature determined in subsequent stages of the work.
