Abstract

This study investigated the morphological, physiological, and biochemical characteristics of Geobacillus sp. B1 isolated from the Rayim So‘fi hot spring in Koson District, Kashkadarya Region, Uzbekistan, and three bacterial isolates, Bacillus sp. M11, Bacillus sp. B, and Bacillus sp. C, obtained from the cotton rhizosphere. The isolates were evaluated for their Gram-staining characteristics, catalase activity, and ability to hydrolyze casein, starch, and lipids. Geobacillus sp. B1 was examined at 50 and 60 °C, whereas the Bacillus sp. isolates were tested at 40, 45, and 55 °C. The results showed that Geobacillus sp. B1, Bacillus sp. B, and Bacillus sp. C exhibited positive reactions for casein and starch hydrolysis, while lipid hydrolysis was not detected in any of the four isolates. In the catalase test, Geobacillus sp. B1 showed a strongly positive reaction, Bacillus sp. B and Bacillus sp. C exhibited positive reactions, and Bacillus sp. M11 showed a negative reaction. These findings demonstrate differences in the enzymatic properties of the bacterial isolates and provide a basis for further investigations into their biotechnological potential.
